If I were to travel to Lion's Arch from Kamadan will I be able to explore and complete all the Tyrian missions and quests or only the missions and quests after LA?

You'll be able to complete all mission's prior to LA, but you won't get any reward for doing them. There are no quest's for you prior to LA however. You'll only start getting bonuses and xp and skill points from mission's after LA.
